چکیده

Transgenic Nicotiana tabacum plants expressing Aspergillus niger b-glucosidase (EC 3.2.1.21) gene (BGL1) in different subcellular compartments [cell wall (Tcw), endoplasmic reticulum (Ter), and vacuole (Tvc)] were analyzed to study the effects of BGL1 localization on plant growth and plant–insect interaction. Transgenic and non-transgenic plants were grown and characterized in a greenhouse with 25/16 8C day/ night temperatures and natural sunlight. Plant insecticidal activity was analyzed with adult whiteflies (Bemisia tabaci) in vial and cage experiments. Compared with wild-type controls, Ter and Tvc transgenic plants did not differ significantly in seed germination, plant growth rate, plant height, or flowering time. However, in Tcw seed germination and beginning of flowering were significantly delayed, and leaf area and plant fresh weight were significantly reduced. Transgenic plants had a marked insecticidal effect on whiteflies (Bemisia tabaci) and on Diptera spp. flies. The density of secretory glandular trichomes was significantly greater in transgenic than in wild-type leaves. This work indicates that hydrolysis of yet to be identified glycosides, may play an important role in plant insect resistance mechanism and plant trichome development. # 2007 Elsevier Ireland Ltd. RNA Interference based Approach to Down Regulate Osmoregulators of Whitefly (Bemisia tabaci): Potential Technology for the Control of Whitefly

Over the past decade RNA interference (RNAi) technology has emerged as a successful tool not only for functional genomics, but in planta expression of short interfering RNAs (siRNAs) that could offer great potential for insect pest management. The diet of insects feeding exclusively on phloem sieves contains water and sugars as main components, and the uptake of the liquid food greatly depends ...

Insecticidal Activity of Some Reducing Sugars Against the Sweet Potato Whitefly, Bemisia tabaci, Biotype B

The effects of 16 sugars (arabinose, cellobiose, fructose, galactose, gentiobiose, glucose, inositol, lactose, maltose, mannitol (a sugar alcohol), mannose, melibiose, ribose, sorbitol, trehalose, and xylose) on sweet potato whitefly Bemisia tabaci (Gennadius) (Hemiptera: Aleyrodidae) survival were determined using in vitro bioassays. Of these sugars, arabinose, mannose, ribose, and xylose were...

Enhanced Whitefly Resistance in Transgenic Tobacco Plants Expressing Double Stranded RNA of v-ATPase A Gene

BACKGROUND Expression of double strand RNA (dsRNA) designed against important insect genes in transgenic plants have been shown to give protection against pests through RNA interference (RNAi), thus opening the way for a new generation of insect-resistant crops. We have earlier compared the efficacy of dsRNAs/siRNAs, against a number of target genes, for interference in growth of whitefly (Bemi...

Mortality of Bemisia tabaci Biotype B (Sternorrhyncha: Aleyrodidae) Adults by Aliphatic and Aromatic Synthetic Sucrose Esters

The B-strain of Bemisia tabaci Gennadius is a key pest of several crops and chemical control is the main control method used by growers, although reduction in efficacy due to insecticide resistance has already been reported. The aim of this work was to investigate the insecticidal effect of an array of synthetic sucrose esters with the aliphatic and aromatic groups on whitefly adults. Sucrose b...

Antioxidant Enzyme Responses Induced by Whiteflies in Tobacco Plants in Defense against Aphids: Catalase May Play a Dominant Role

BACKGROUND Bemisia tabaci MEAM1 (Middle East-Asia Minor 1) feeding alters antioxidative enzyme activity in some plant species. Infestation of B. tabaci nymphs decreases Myzus persicae performance on systemic, but not local leaves of tobacco plants. However, it is unclear if B. tabaci nymphs induced antioxidant activities contributing to the aphid resistance. METHODOLOGY/PRINCIPAL FINDINGS We ...
